Skipper Asghar Afghan struck 49 off 28 balls to lift the team to 184-4 after winning the toss in Greater Noida in India.
In reply, Ireland could only manage 163-6 off 20 overs.
Afghan spinner Mujeeb Ur Rahman took 3-38 to earn the man of the match award.
The third and final game of the series will be played at the same venue on Tuesday.
